The dysconnection hypothesis states that schizophrenia is associated with reduced synaptic connectivity (in which case it is sometimes spelt ‘disconnection’ – lack of connectivity) or dysfunctional connectivity (i.e. dysconnection – abnormal functional connectivity) primarily in the mesocortical pathway (i.e. midbrain dopamine and serotonin afferent to the PFC) and between cortical areas such as the frontal cortex and the temporal lobes.
